Gene Synthesis> | For InDel analyses, 10 ng of each DNA sample were used for each 25 μL reaction with 2× reaction Taq buffer (500 mM KCl, 100 mM Tris-HCl, 15 mM MgCl2, Triton x-100), 1 μL of each primer (0.4 μM), 2.5 mM of each dNTP, and 0.2 units of Taq DNA polymerase (GenScript USA Inc., Piscataway, NJ, USA) per reaction. Amplifications were performed using a PTC-100 Programmable Thermal Controller (MJ Research, Inc., Waltham, MA) programmed with the following conditions: temperature of 94 °C for 2 min, followed by 35 cycles of 94 °C for 30 s, 52 °C for 30 s, and 72 °C for 1 min; with a final extension at 72 °C for 7 min. | Get A Quote |
With increasing use, manufactured nanomaterials (MNMs) may enter soils and impact agriculture. Herein, soybean (Glycine max) was grown in soil amended with either nano-CeO2 (0.1, 0.5, or 1.0 g kg−1 soil) or nano-ZnO (0.05, 0.1, or 0.5 g kg−1 soil). Leaf chlorosis, necrosis, and photosystem II (PSII) quantum efficiency were monitored during plant growth. Seed protein and protein carbonyl, plus leaf chlorophyll, reactive oxygen species (ROS), lipid peroxidation, and genotoxicity were measured for plants at harvest. Neither PSII quantum efficiency, seed protein, nor protein carbonyl indicated negative MNM effects. However, increased ROS, lipid peroxidation, and visible damage, along with decreased total chlo...
